If you are all on the same booking, you'll have to check-in together.

What exactly does "checking in together" mean? There are 45 of us on our booking.....what would be expected of us when we get to the desks?

It means you all have to check-in together. Because all passengers are listed under the same booking, you'll all have to be present to check-in at the same time. The passenger names, passports and identity have to be matched. You can't check-in a few at a time seperately. Refer to the booking conditions for the respective airline and their check-in procedures. For example, Thomas Cook's.
