We travelled down the Turnpike when we went from Orlando several years ago.There are rest areas every few miles in the centre of the carriageway.
This road was less busy than the main highway which I think is the A1.

Rental car is the only viable way and takes about 4 to 5 hours via the Turnpike.

Why not just fly into Miami? It's only 25 miles to Port Everglades at Fort Lauderdale.
If flights to Orlando work better just rent a car and drive down in around 4 hrs. Do NOT take the A1A road, it is an old road than runs all the way down the coast, you go through town after town..
