Why not ask them?

Put it that due to work/family problems a couple may not be able to go and how will it effect the booking.

If there are cancellation charges then rather ask now they are low than wait.

Your under occupancy supplements will change and probably increase the cost of the holiday. That means that the 5 remaining travellers will have to pay more.

The other alternative is to find two people to take their place and get the two who are cancelling to cover the admin fees for changing the names.

As suggested by fwh you can ask them now, without doing anything and then check your options, however, if you do decide to change the names on the booking you can leave this up to about 10/12 weeks before departure, but please bear in mind that if you do remove them off the booking then then your costs will most definitely increase.
